Santana's relationship with her parents is largely unknown, apart from the fact that her dad is a hot-shot doctor with an apparently smokin' insurance plan. Santana obviously wants for nothing; she was even allowed to have a breast enhancement surgery during the summer between seasons 1 and 2.
She's a member of the Cheerios, Sue Sylvester's elite cheerleading squad at McKinley High School. She is granted the role of Head Cheerleader once Quinn becomes pregnant and kicked off the squad during season 1, but loses this role back to Quinn in season 2 when Coach Sylvester finds out she had breast enhancement surgery over the summer. Santana jumps Quinn in the hall next time she sees her, seeking to return the humiliation by attacking her physically and calling her out over her previous pregnancy.
During the beginning of season 1, she was also a member of the Celibacy Club; like most of the other members, however, her membership was intensely hypocritical.
She joins Glee Club along with her best friend and fellow Cheerio Brittany S. Pierce only because Coach Sylvester decides they would make the perfect pair of spies to aid her with her plan of destroying the Glee Club. She truly comes to love Glee Club however, branding it as the "best part of her day."
Santana is mostly shown as being promiscuous: sexting with Puck, taking Finn's virginity, apparently having sex with Brittany, and sleeping around with guys in general. At one point she and Puck were actually dating, but she broke up with him due to his low credit score. Despite having dumped him, she grows very jealous of fellow Glee Clubber Mercedes when she later "dates" Puck; things get physical between the two girls when they perform a very tense and loaded version of "The Boy is Mine."
When everyone else falls for Jessie St. James' switch over from rival Vocal Adrenaline to McKinley's New Directions, she is the only one to see through this and consider him as a spy for the other side.
Santana is obsessed with her image and becoming an "undisputed top bitch" of McKinley High; this obsession heavily underscores a good majority of what we see from her but does not wholly define her character.
She has no qualms with bullying others that she considers beneath her in some way, a fact which mutes in intensity somewhat as she becomes more involved with Glee Club but has yet to completely go away. She is verbally sassy, utilizing both well-timed wit and ghetto slang to ramp up her intimidation factor. She's extremely self-entitled, shown to behave territorially when anyone encroaches upon what she perceives as rightfully 'hers' (Puck, Brittany, the BreadstiX gift certificate, her status as Head Cheerleader, etc.).
She's also shown to be an active social-climber, "dumping" her unofficial boyfriend/casual sex partner Puck due to his low credit score, but this has been shown to backfire on her on at least one occasion. Specifically during Duets, her close relationship with her best friend and fellow Cheerio Brittany S. Pierce is substantially threatened when Santana flies into "gay panic" mode after Brittany suggests partnering up to sing a duet to Melissa Etheridge's song "Come to my Window." Prior to this, Santana has never been shown to be anything but affectionate and kind towards Brittany, even after the pair of them were outed by Brittany during Sectionals as having had sex before, so Santana's cold-hearted remarks to Brittany that she's only there right then because Puck's stuck in juvie, that she's like a snake that needs something warm underneath her in order to digest her food, as well as Brittany's (understandably) poor reaction to these statements, leave reasonable cause to assume that Santana is more into Brittany than she is willing to let on but unable to let things progress to such an obvious and in-your-face status that might cause serious damage to her reputation in homophobic Lima, Ohio. More reason to doubt Santana's vehement denial in any actual romantic feelings toward Brittany come during the same episode, where Santana is shown to be visibly bothered by Brittany getting along without her constant company. She also exhibits outright jealousy by successfully acting to break up Brittany and Artie's blooming relationship; whether this can simply be attributed to her possession issues or whether her petty behavior is actually indicative of more mature feelings toward Brittany is currently unknown however. During the following episode, The Rocky Horror Glee Show, Santana and Brittany seem back to being their old selves around each other without any explanation. This might be touched upon again in future episodes (a thought that's supported by actual cast member speculation) but for now the true nature of their relationship remains unknown.
Also worth noting in this section is the fact that Santana has no problem following Coach Sylvester's strict Master Cleanse regimen - a mixture of water, maple syrup (for glucose), lemon (for acid), cayenne pepper (to irritate the bowels), and ipecac (a vomiting agent). This is all done in order to keep her figure and weight down for the sake of cheerleading, and Santana's easy submission to such a harmful practice is yet another example of Santana's tendency to do anything for the sake of preserving her status around McKinley high.
For a girl so fixated on cultivating such a tough image, the fact that she breaks down in near tears while serenading Mr. Schue with lyrics like "A friend who taught me right from wrong / and weak from strong / that's a lot to learn" during Glee's rendition of "To Sir, With Love," is very telling; she's not all thorn and thistle. Actually, her smarmy air of confidence is disturbed quite often, exposing a vulnerable side to her character. Sometimes it's over petty things, like losing tanning privileges or being denied a free meal at Breadstix. But most troubling of all her quirks may be why such a pretty and popular girl felt the need to undergo breast augmentation. At first she appears fairly pleased with herself - sassier and possibly more promiscuous than ever (if the reaction of McKinley's male population is anything to go by). But she faces harsh criticism from rival Quinn Fabray and - more seriously - Coach Sylvester, who actually demotes her from her position as Head Cheerleader and continues to openly mock her over her "vine-ripened chest fruit." Santana even catches flack from the increasingly chauvinistic Artie, who wonders if he can use the ostrich eggs she's smuggling in her bra to make the world's largest omelette.
Overall, Santana is shown to have a very wholesome devotion to Glee Club; she describes it as being the best part of her day (although she specifically instructs everyone to maintain that she hates it, if asked). She's very downtrodden just prior to their performances at Regionals, skeptical of their ability to defeat rival club Vocal Adrenaline and requiring extra encouragement to get her 'game face' on. She's obviously worried about the possibility of having Glee taken away from her, building on her previous admission of Glee being the best part of her day.
This all, along with her patient indulgence of Brittany's outlandish naïvety and their frequent cutesy physical proximity (hugging, cuddling, walking down the hallways with their pinkies linked, etc.), speaks of a very complex character in spite of her seemingly 2D "mean girl cheerleader" setup. She's got a hardened exterior but she's surprisingly tender inside and it's this strange duality that gives her character such a good deal of charm.
- Santana learns how to effectively shop lift from April Rhodes.
- She disapprovingly considers bakesales "bougie" despite being rather "bougie" herself.
- Ranks 2nd place on the "Glist" (after Quinn).
- "Wants" to "get her anesthesia on" at the dentist despite having no real need (perfect teeth); shares a drug-induced dream sequence with Brittany, in which she plays Madonna to the other's portrayal of her iconic namesake.
- Has a tentative relationship with Mercedes Jones. The pair fought over Puck and still don't particularly like one another, but they both recognize each others vocal prowess and how well their voices compliment one another. Santana specifically sought out Mercedes to do a duet to "River Deep, Mountain High" even after rejecting Brittany.
- Was a voyeur along with Brittany outside the window of Ms. Pillsbury's office when she and Mr. Schue got up to a sexually charged practice session of "Touch-a, Touch-a, Touch-a, Touch Me."

Santana Lopez in the house. All you mouth-breathers better seriously appreciate what I'm doing for you right here. There's precious little time left leading up to Nationals now and if Coach Sylvester finds out I'm here giving this stupid interview instead of getting ready for our Saturday practice session she'll kick my ass right out of my spankies. So, chop-chop. Vámonos. Let's get this freak show on the road. You get ten minutes to ask all your burning questions. But let's be perfectly clear on one thing: we do not talk about Glee. At all. Whatsoever. Got it? Good.
Third Person:
Her hips move with provocative purpose; the swish of her pleated uniform draws attention as intended, to the shapely plain of her outer thigh. Santana smiles, white and pearly, at the meathead whose lewd fantasy she surely now occupies. "Come on," she purrs, fingertips following down the grain of his bicep, as she swaps standing in front of him for standing behind. "Let's get out of here for a bit."
In twenty minutes' time, they're squeezed into opposite sides of a booth at Breadstix. Santana's smile has changed from predatory to placated. Karofsky appears mildly disgruntled in return, as if recognizing somewhere in his little pea-sized brain that he's been duped. But the sexual frustration melts right off his face as the waitress arrives with their first basket piled high with warm, garlic-y deliciousness.
Santana reaches out to grab one between two perfectly manicured nails, blowing lightly on the end before taking a bite. The whole "better than sex" cliché is way overused, but she considers these staples of modern Italian cuisine to be nothing short of orgasmic.
After all, she doesn't fool around with Karofsky for his tiny dick or 'charming' personality. He's one of the only guys she knows with a bottomless wallet and a truck with a bed capable of hauling off all the excess breadsticks that the restaurant is legally forbidden to stop serving her.
